Molecular target

BCR-ABL Tyrosine Kinase

Constitutively-active oncogenic fusion kinase produced by the Philadelphia chromosome (t(9;22)) in chronic myeloid leukemia. Selective inhibition by imatinib (Gleevec — first targeted therapy approved 2001) transformed CML from rapidly fatal to a manageable chronic disease. Subsequent kinase inhibitors (dasatinib, nilotinib) target the same lesion.
